<para>This study investigated patterns of nitrogen (N) fixation rates, leaf N content, and geographic diversity in the N-fixing bacterium Frankia occurring in symbiosis with Alnus incana ssp. tenuifolia and A. viridis ssp. fruticosa in early and late successional habitats on the Tanana river floodplain and surrounding uplands in the Bonanza Creek Experimental Forest. Frankia diversity was estimated via polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) of the nifD-K spacer region, a non-coding region in the nitrogenase-encoding operon. Specific N-fixation rate was measured with a 15N2 uptake assay and leaf N content via mass spectrometry. Additional parameters measured were soil temperature and moisture, leaf del 15N, and specific leaf area. </para>

<section>
<title>Experimental Design</title>
<para>At each site 10 alder plants were chosen haphazardly; at late succession floodplain sites in which both host species co-occurred 10 plants of each species were chosen. Each site was sampled three times across the growing season, with different plants chosen at each sampling period. Approximately 2.5 grams of nodule tissue was harvested from each plant and assayed for N-fixation rate using a 15N2 uptake assay. Additionally, at each plant soil temperature was measured at 1 cm and 5 cm depths, a soil core was taken for gravimetric moisture measurement, and five leaves were chosen for measurement of specific leaf area, leaf N content, and del 15N. Half of the 15N-assayed nodule sample was selected for DNA extraction and selective amplification of the Frankia nifD-K non-coding spacer locus via polymerase chain reaction (PCR). PCR product was probed for sequence variation by digestion with the restriction enzymes CfoI and HaeIII, followed by high resolution gel electrophoresis. 

Data were entered and double-checked by Mike Anderson. Soil moisture was calculated as weight of oven-dried soil cores divided by the corresponding fresh weight, times 100. Calculation of specific N-fixation rate (SNF) follows the methods in Anderson et al., 2004. Leaf N content and del 15N were measured via mass spectrometry. Specific leaf area was obtained by dividing dry mass of circular punches taken from leaves by the total area of the punches.
