<para>Prior to this investigation, denitrification had not been intensively measured in headwater catchments of interior Alaska.  The primary purpose of this database was to estimate the importance of denitrification as a mechanism of nitrogen retention in the riparian zone of watersheds underlain by discontinuous permafrost.</para>

<section>
<title>Experimental Design</title>
<para> Denitrification rate was measured on riparian soils on five dates from May 25 and September 1, 2004 using the acetylene block technique on intact soil cores constructed of ABS pipe (3.8 cm ID x 25 cm length).   Soil cores were collected from randomly-selected points to a depth of 15 cm in the valley bottom of each watershed (25 to 30 cores per watershed per date).</para>

<title>Methods</title>
<para>Field Methods:  After collecting soil, cores were capped with rubber stoppers.  One stopper was modified with a sampling port for sampling gas in the headspace.  After capping with rubber stoppers, cores were amended with 50 ml of stream water and 20 ml of acetylene (10% by total volume), and then shaken vigorously to aid equilibration between aqueous and gaseous phases.  Cores were stored in a cooler to maintain the temperature near in situ conditions during the incubation (2 - 6 ºC).  Gas samples for nitrous oxide measurement were collected at 2 (initial) and 6 hours (final) following the acetylene addition with a 60 ml syringe and transferred to an evacuated serum vial until analysis.  Lab methods:	Nitrous oxide concentration was measured using a Varian CP-3800 gas chromatograph equipped with an electron capture detector.  Following incubations, soil was dried at 95 ºC in order to calculate denitrification on a soil dry mass basis.  Processing procedures:	Denitrification rate was calculated as the difference between final and initial nitrous oxide concentration (corrected for gas volume in headspace, soil water volume, and nitrous oxide dissolved in aqueous phase, Bunsen coefficient = 0.02 moles N2O L-1 H2O). </para>
